A cell map is not yet evidence of circuit repair

Research report · 14 October 2026 · Cell states and functional repair

Publication note: Research assessed on 14 October 2026; published with a delay on 15 October 2026. Read the subsequent assessment of 15 October.

Which link between new cells and recovered function has actually been measured?

Project status: 🔵 Level 2 · AG-C06; AG-T01 unchanged. Evidence: moderate descriptive animal-model evidence; ⚪ Literature Level 1. No established reversal of aging.

Today’s refinement

The deeper main-text assessment of Wei 2022 strengthens the cellular component of axolotl regeneration. It does not yet verify a direct counterfinding to Amamoto’s circuit deficits. Both studies can be correct at different measurement levels.

Comparison point Verified finding
Time and site Wei: 2–60 days after injury; cites Amamoto’s protocol. Lateral/dorsal site descriptions differ within the paper. Exact lesion equivalence unresolved.
Biological unit 15DPI–1 through –4 are four sections from one animal. Complete animal counts, age and sex unverified here.
Measurement and inference Spatial gene expression and cell markers; RNA velocity, pseudotime and ligand–receptor predictions. Comparable tract, circuit or behavioral recovery not verified in the examined material.

Decision and alternative explanations

Project inference: A cell map addresses different questions from measurements of functional connections. Computed state transitions or communication signals must therefore not be treated as direct lineage tracing, axon tracing or signal transmission. Four sections improve spatial resolution but do not replace four independently studied animals. That allocation alone does not prove a flaw in the original statistics.

Yesterday’s Amamoto assessment concerned physiology at 11–15 weeks. A valid comparison needs appropriate time windows, injuries, controls and functional endpoints. Differences could reflect maturation, lesion extent or different measurement targets; none of these explanations is confirmed here. Function that was not assessed remains open and is not a negative functional test. AG-C06 remains Level 2. No new efficacy evidence follows for TIMP2, Hand2 or human aging.

The next executable test

The original article lists ARTISTA as a code resource. Next, only its README and small explicitly linked sample metadata will be checked for animal/section mapping. No large matrices or invented identifiers. This route was not opened today. A functional reassessment requires an explicit connectivity or circuit assay with control, timing, experimental unit and uncertainty; another cell map alone would not close this gap.

Evidence limits and sources

Established within this assessment: the explicitly read details above. Promising signal: cellular reconstruction. Experimental results: external spatial-expression/marker findings. Hypothesis: ensuing functional integration, still untested. No project experiments, effect calculation or independent reproduction.
