New neurons are only the beginning

Research report · 13 October 2026 · Regeneration and functional integration
When does a new neuron become a functioning part of the brain again?

Project status: 🔵 Level 2 · AG-C06; AG-T01 unchanged. Scientific evidence: moderate preclinical evidence from a peer-reviewed injury study. ⚪ Literature Level 1. No evidence of reversal of aging.

The decisive counterexample

Amamoto and colleagues examined regeneration following mechanical injury to the adult axolotl pallium in 2016. Regenerated neurons showed electrical activity and local synaptic inputs, while examined connections and circuit responses remained impaired. Producing new cells alone is therefore insufficient evidence of complete functional repair. This finding is new to the project registry today, not new to science. Original study · 9 May 2016

Assessment level Finding and limit
Cell function Patch-clamp at 11–15 weeks after injury: 14 regenerated and 9 control neurons. Electrical activity and local inputs. Neurons are not independent animals; corresponding animal n unverified here.
Architecture and connections Altered cellular organization at 11 and 20 weeks; tracing/MRI show limitations of examined distant connections. Not evidence of lifelong failure to recover or complete loss of function.
Circuit response Reduced field responses at 11–15 weeks after injury. Dorsal→lateral: 5 control/3 injured animals, 15 slices per group. Dorsal→medial: 4/3 animals, 9 slices per group. Do not add slices and projections as independent animals or a pooled effect.

Original passages and figure legends 4–7. No project-derived effect size, uncertainty estimate or raw-data analysis.

What changes in the target model

Project synthesis: For neuronal replacement, AG-C06 requires four separate lines of evidence: appropriate cell identity, connections to the right targets, functional circuit responses and sufficient durability. If a predefined functional success criterion is reliably unmet after an appropriate interval despite successful cellular reconstruction, this argues against complete repair in the examined model. Function that was not measured instead leaves the claim open and unsubstantiated. It would not erase the biological partial advance.

Alternative models include delayed integration, different compensating circuits and differences in injury extent. A smaller field response may reflect fewer activated axons or altered excitability; it does not alone identify a specific synaptic mechanism. Behavior and age-related functional loss were not resolved by today’s assessment. For the approaches registry, this is a separate brain countercontext to spatial regeneration control, not a shared Hand2 mechanism. It neither confirms nor refutes TIMP2.

Next executable step: Inspect the already registered spatial single-cell study AG-S02 (Wei et al., 2 September 2022, DOI 10.1126/science.abp9444) specifically for connectivity and functional measurements. Distinguish cell states, tracing and physiology; compare injury site, extent, timing and experimental units. This comparison has not yet been performed. A future external confirmation test would need to link cellular reconstruction and defined function within the same model; this project cannot conduct laboratory experiments.

Evidence, correction and teaching

Established within this limited assessment: the reported statements in selected original passages. Promising signal: locally functional neuronal regeneration. Experimental result: the model-specific dissociation between cellular and circuit endpoints. Hypothesis: coordinated restoration of cell identity and connectivity could enable functional repair; no established treatment of human aging. AG-C06 remains Level 2; AG-T01 remains Level 2 with low mechanistic confidence.

Correction to 12 October: The institutional AG-D06 PDF locator was already known on 7/8 October. Its text was accessible again today, without a new substantive finding or verified supplement. The description as a new locator is withdrawn; TIMP2 sensitivity gaps remain open.

Some direct requests limited by challenges/retrieval errors. Targeted DOI search found no attributable correction/retraction; not a complete exclusion check. +1 historical original study: 29 studies, 28 peer-reviewed and one preprint. Search and limitations.

The existing professor module “Regenerated neurons and restored function” already covers this question; no duplicate wish. AG-P21 of 11 October remains unpublished pending the native contract review. Research colloquium · Evidence chain · Approaches registry.
