Removing senescent cells: which cells, which benefit?

Research report · 17 October 2026 · Counterevidence and cell selection

'''Guiding question: Which cell population is actually removed — and which comparison identifies its contribution to repair?'''

'''Today's decision:''' “Early removal harms, later removal helps” is not justified as a general rule. AG-C06 gains counterevidence and a more precise control comparison. The discrepancy remains unresolved; one possible explanation can now be tested more clearly.

== Counterevidence admitted · AG-D09 ==
[https://doi.org/10.1038/s43587-024-00755-4 Gasek et al., Nature Aging: Clearance of p21 highly expressing senescent cells accelerates cutaneous wound healing]. Online '''13 November 2024''', January 2025 issue; assessed '''17 October 2026'''. Previously published, newly registered today. Access: indexed original passages from the [https://pmc.ncbi.nlm.nih.gov/articles/PMC12205026/ PMC author manuscript], not another study or preprint.

p21-high/DTA mice, 8–12 weeks; both genotype groups receive tamoxifen. Improved closure reported in females, no significant benefit in males. Two 6-mm wounds per mouse; n denotes mice. Methods report no randomization or blinding. A tamoxifen/vehicle control exists. Wound aggregation and effect confidence intervals not verified here, limiting confidence in the comparison.

== Prior knowledge and countermodels ==
Yesterday's AG-D08 reports impaired healing after early p21 interventions. Its discussion already explicitly addresses AG-D09: p21-high selection, different killing mechanisms and tamoxifen regimens are proposed explanations. These are existing literature proposals, not established explanations. AG-D08 uses FKBP/caspase-8 activation for cell removal; DTA inhibits protein synthesis. Model names alone do not fully establish which cell populations differ. [https://www.nature.com/articles/s41556-026-01948-2 AG-D08 discussion, online 28 May 2026; specifically rechecked today]. ! Assessment decision !! Reason and remaining gap
|-
| Do not isolate timing from this comparison
| Comparing studies changes several conditions together. It does not estimate a pure timing effect.
|-
| Equal drug exposure is an important control
| With equal exposure and an additive model, a shared tamoxifen main effect alone cannot explain a genotype-group difference. Genotype-dependent effects remain possible.
|-
| Require direct contrasts
| “Significant in females, nonsignificant in males” does not establish a sex difference. Multiple wounds and time points require documented mapping to animals.
|}

== Next project test: what can actually be calculated? ==
The functional comparison first requires '''Δ = mean wound closure in the intervention group minus mean wound closure in the control group''', using the same normalization and prespecified time point. For percentage closure, the unit is percentage points. Define C(t) = 100 × [1 − A(t)/A(0)] for closure and ΔC = C(intervention) − C(control); A is area in the same unit, t the shared measurement time. Positive ΔC means more closure at that time, not a measured speed parameter. Remaining normalized wound area has the opposite sign. This is not general rejuvenation. This is an analysis specification, not a calculated result.

'''Next executable step:''' Inspect the small original-linked source-data workbook for Fig. 2 / Extended Data 7 for animal ID, sex, genotype, time, starting area and aggregation of both wounds. Calculate direct group differences and uncertainty only if units are traceable; assess sex differences through a direct contrast. Otherwise record the specific missing mapping. Do not pool with AG-D08 or treat reconstructed bars as raw data.

'''Counterhypothesis and falsification:''' A selection model must explain a functional difference after accounting for schedule, genotype, exposure and measurement. Assessment after correct unit mapping considers the effect estimate and precision; loss of nominal significance alone does not refute benefit. External experimental confirmation would need direct measurement of target cells and actual elimination; none was performed here. Cell markers, measurement methods and subanalyses of one paper do not count as independent replications.

== Evidence and project status ==
* '''Established knowledge:''' Both publications exist and address different intervention contexts; bibliographic and retrieval dates are recorded separately.
* '''Experimental findings:''' Animal wound healing. AG-D09 is admitted as limited preclinical evidence with bias concerns, not human efficacy.
* '''Promising indication:''' More precise cell selection might separate repair from harmful states; benefit remains untested.
* '''Hypothesis / project inference:''' The cause of the divergent healing effects has not been identified. Control contrasts and target-cell verification take priority over a mechanistic explanation.

⚪ '''Level 1:''' AG-D08/AG-D09 literature and approach AG-A08. 🔵 '''Level 2:''' AG-C06 synthesis; AG-T01 unchanged with low mechanistic confidence. No Levels 3–5 or independent confirmation of an aging solution. This supplies an assessment criterion for axolotl and brain research, not a shared p21/TIMP2 mechanism. ARTISTA and other parked access routes were not repeated.

== Search and teaching ==
Targeted counterevidence appraisal, DOI/date checks and search for attributable corrections/retractions; none found, not a complete notice audit. Direct publisher/PMC access partly limited; selected indexed original passages accessible. No complete supplement, raw-data or current-literature audit. Source delta +1: '''31 DOI-distinct papers''', 30 peer reviewed and 1 preprint; 12 approaches, 6 claims.
